I found the forth one... still clueless.... Does it relate to the names of the pieces of the chess?
After thousands of times of attempts trying to build relationship(e.g. mirror image) between the two chessboards, I must tell all the newcomers: Don't care about the wrong positons of dark&light squares, and even no need to think about the white&dark chessmen.
I would apologize if the comment above spoil too much. Even though, I must say this level bewilders and misleads people by involving chess rules which turn out to be completely useless.
Gamemaster, I expected something more ingenious and interesting just like lvl.19,31, etc..I hate enumerating all the cases(e.g. lvl.28) and don't like the vagueness of this level.

Do I need to reverse the second board upsidedown after filling it with letters?

Not necessarily needed. You can rotate that if you are confused. Compare and see what is similar.

Do I still need the four clues for the "similarity" or the final answer?
Or just the two boards are enough?

No, the four clues are indeed used only to find the pattern. Nothing else.
